Recognizes a protein of 55 kDa, which is identified as fascin-1. Its actin binding ability is regulated by phosphorylation. Antibody to fascin-1 is a very sensitive marker for Reed-Sternberg cells and variants in nodular sclerosis, mixed cellularity, and lymphocyte depletion Hodgkin's disease. It is uniformly negative in lymphoid cells, plasma cells, and myeloid cells. Fascin-1 is also expressed in dendritic cells. This marker may be helpful to distinguish between Hodgkin lymphoma and non-Hodgkin lymphoma in difficult cases. Also, the lack of expression of fascin-1 in the neoplastic follicles in follicular lymphoma may be helpful in distinguishing these lymphomas from reactive follicular hyperplasia in which the number of follicular dendritic cells is normal or increased. Recognizes a glycoprotein of 44-88 kDa, which is identified as CD28. It is the critical T-cell co-stimulatory receptor which provides to the cell the important second activation signal by binding CD80 and CD86 that are expressed by antigen presenting cells. Besides its co-stimulation role, CD28 functions in preventing T-cells from anergic hyporesponsive state or from undergoing premature apoptotic cell death. CD2 interacts through its amino-terminal domain with the extracellular domain of CD58 (also designated CD2 ligand) to mediate cell adhesion. CD2/CD58 binding can enhance antigen-specific T cell activation. CD2 is a transmembrane glycoprotein that is expressed on peripheral blood T lymphocytes, NK cells and thymocytes. CD58 is a heavily glycosylated protein with a broad tissue distribution in hematopoietic and other cells, including endothelium. This MAb reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40 kDa. Its epitope maps between amino acid 312-335. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. The highly leukemogenic avian retrovirus E26 contains two oncogenes, v-Myb and v-Ets, which are expressed together as a fusion protein. The cellular homolog of v-Myb, designated c-Myb, encodes a transcription factor. This antibody recognizes a carbohydrate epitope on a single chain, transmembrane, heavily glycosylated protein of 90-120 kDa, which is identified as CD34 (VI international workshop on human differentiation antigens). Its expression is a hallmark for identifying pluripotent hematopoietic stem or progenitor cells. Its expression is gradually lost as lineage committed progenitors differentiate. CD34 is a marker of choice for staining blasts in acute myeloid leukemia. In addition, it is expressed by soft tissue tumors, such as solitary fibrous tumor and gastrointestinal stromal tumor. CD34 expression is also found in vascular endothelium. Additionally, proliferating endothelial cells overexpress this molecule than the non-proliferating endothelial cells.
